چکیده

Posidonia oceanica is an endemic seagrass species in the Mediterranean Sea. Due to its ecological importance, it has been the subject of several genetic studies aimed at increasing general knowledge of the system and helping to define appropriate management strategies. We compared the efficiency of 2 different classes of molecular markers, ‘inter simple sequence repeats’ (ISSRs) and ‘simple sequence repeats’ (SSRs) or ‘microsatellites’, in population genetic analysis of P. oceanica. To do this, we analyzed meadows along the coasts of the Calabria peninsula (Italy, South Tyrrhenian Sea and Ionian Sea), a putative biogeographic barrier for P. oceanica. SSR and ISSR markers detected different levels of genetic variability within P. oceanica populations. Dinucleotide microsatellites were more polymorphic than triand heptanucleotide microsatellites and ISSRs. Nevertheless, discriminating power analysis performed for each single locus showed that some trinucleotide microsatellite and ISSR primers were more sensitive than dinucleotides. The use of selected ISSR and SSR markers together allowed the detection of higher polymorphism than either set of marker alone. Tyrrhenian meadows were always more polymorphic than Ionian ones, according to both SSRs and ISSRs. Both markers strongly suggested the Calabria peninsula as an important biogeographic barrier between the Western and the Eastern side of the Mediterranean basin.

Transcriptome characterisation and simple sequence repeat marker discovery in the seagrass Posidonia oceanica

Posidonia oceanica is an endemic seagrass in the Mediterranean Sea, where it provides important ecosystem services and sustains a rich and diverse ecosystem. P. oceanica meadows extend from the surface to 40 meters depth. With the aim of boosting research in this iconic species, we generated a comprehensive RNA-Seq data set for P. oceanica by sequencing specimens collected at two depths and two...

Comparative Analysis of Stability—Genetic Diversity in Seagrass (Posidonia oceanica) Meadows Yields Unexpected Results

The diversity–stability relationship is the subject of a long-standing debate in ecology, but the genetic component of diversity has seldom been explored. In this study, we analyzed the interplay between genetic diversity and demographic responses to environmental pressures. This analysis included 30 meadows formed by the Mediterranean endemic seagrass, Posidonia oceanica, showing a wide range ...

Comparison of ISSR and AFLP markers in assessing genetic diversity among Nettle (Urtica dioica L.) populations.

Urtica dioica is an important medicinal plant which is widely distributed in Mazandaran province (North of Iran). In this study for the first time Amplified Fragment Length Polymorphism (AFLP) and Inter-simple Sequence Repeat (ISSR) markers were used for detection of genetic polymorphism in Mazandaran nettle. Ten AFLP primer combinations and seventeen ISSR markers were utilized. AFLP produced 830...

Genetic Diversity Evaluation of Lemon balm (Melissa officinalis L.) Ecotypes Using Morphological Traits and Molecular Markers

Genetic diversity of 12 lemon balm ecotypes was evaluated using agronomic traits as well as ISSR and RAPD markers. Results of analysis of variance showed high diversity among the studied ecotypes. Cluster analysis using UPGMA method grouped the ecotypes into three different groups based on the agronomic traits. Twelve ISSR primers created 106 polymorphic bands among the studied ecotypes. An ISS...

Genetic diversity analysis of recombinant inbred lines of rice (Oryza sativa L.) using microsatellite markers

Estimation of genetic diversity is an important factor in germplasm conservation and characterization. In rice breeding programs, genetic diversity information on specific regions of genome can be very useful for the application of marker assisted selection (MAS) and for gene mapping. A total of 152 rice lines were considered for breeding programs using microsatellites (SSR) technique. The tota...
